課程概述

Goal of this course is to show how optical methods can make a difference in our understanding of significant biology problems. Topics such as ligand-DNA interaction, cancer biology, cell tracking and molecular motors are introduced. We hope to introduce the significant biological principles involved and the range of optical methods that can be employed to produce new knowledge about these topics at a fundamental level.

(1). Introduction to optical methods (Chang and Li) 12-18 hrs
- basic spectroscopy (introduction of laser, group theory and vibrational spectroscopy), various spectroscopic methods
- basic microscopy (introduction to various imaging, tracking methods and their applications in biological sciences)

(2). Non-linear spectroscopy (Shen) 4 hrs

(3). Novel spectroscopy and microscopy approaches to Biology (Yeung) 4 hrs

(4). Theory (Lin) 4 hrs

(5). Research projects (8 half-days in the research lab)

Research projects: As part of the course, students are required to engage a research project, in a group of 5 people, which uses the materials learned from this course to address a biological question. The project is a required part of the course, and will take 50% of the final grade.
(i). Single molecule method
single molecule TPM method/magnetic tweezers technique to study protein-DNA interaction
(ii). Spectroscopy Combination
Absorption, fluorescence and CD spectroscopy in studying DNA topology
(iii). Raman, CARS and SERS spectroscopy/microscopy
Non-linear spectroscopy for imaging
(iv). fluorescence lifetime, FLIM, FRET methods
Fluorescence-based method for study ligand-DNA interaction
